4-(3-oxobut-1-enyl)benzoylated hydrophilic support(301)(scheme 65)[310]:To 4-(3-oxobut-1-enyl)benzoic acid (85mg,0.45mmol) and N-ethyl maleimide (NEM)(95μL,0.75mmol)dissolved in DMF (2ml) was added o-(7-azabenzotriazol -1-yl)-1,1,3,3-tetramethyluronium hexafluorophosphate(HATU)(160mg,0.42mmol)and 1-hydroxy -7-zazbenzotriazole(HOA他）（20mg,0.15mmol).The reaction mixture was allowed to pre-activate for 5 min at ambient temperature prior to addition to resin(298)(500mg,0.3mmolg-1)with a free amino group.The reaction was allowed to continue until a negative Kaiser test was obtained.
(The NEM was added to sequester excess coupling reagent in order to avoid side products.) The reaction solution was drained and the resin was washed as follows:
1.5*10mL DMF
2.5*10Ml CH2CL2
The resin was then dried in vacuo.
